Thus, a grain was ground, and approx. 20 mg was suspended in 0.5N NaHCO3, mixed with EtOH, centrifuged, the supernatant decanted, and the residue mixed with 3 mL dansyl chloride soln. (1 mg/mL EtOH) and allowed to stand 1 h. The product was washed with aq. EtOH suspended in H2O, and measured fluorometrically. Fluorescence intensity was linear with concn. up to 25 mg flour/5 mL, and was proportional to the amt. of lysine [56-87-1] in the sample.
