The exts. from the hearts of adult hens and 14-day-old embryos displayed a single peak of I activity at a slightly different elution vol.Several substances with their cas registry numbers 61-19-8 and 2140-58-1 may be metioned in this study., whereas in the heart ext. of 1-day-old chickens 2 mol. forms of I were eluted. The kinetic and regulatory properties of the purified adult hen heart I were studied and compared with those of the corresponding enzyme from 14-day-old embryo hearts. Both enzymes exhibited a slightly sigmoid-shaped plot of the reaction rate vs. substrate concn., which shifted to hyperbolic form when ATP or ADP was added into the incubation medium. The enzymes were strongly activated by ATP, less efficiently by ADP, and the activating effect was enhanced at low substrate concn. Orthophosphate inhibited both enzymes but this inhibition was more potent for embryo heart I. Palmitoyl-CoA inhibited adult hen but not embryo heart I. Thus, differences in the regulatory properties of the mol. forms studied do exist and they correspond with the ontogenetic differences obsd. previously concerning the effect of temp. on chicken heart I. .
