DOCUMENT TYPE: Journal CA Section: 1 (Pharmacology) The therapeutic efficacies of the newer b-lactam antibiotics piperacillin [61477-96-1], azlocillin [37091-66-0] and aztreonam [78110-38-0] were compared with the efficacies of ticarcillin and tobramycin in a guinea pig model of exptl. P. aeruginosa pneumonia. For animals challenged with 2 ′ 108 CFU(colony-forming unit), tobramycin treatment resulted in survival rates and intrapulmonary killing of pseudomonads which were significantly greater than those found with any of the b-lactam agents. There were no significant differences noted among the individual b-lactam agents. When animals were challenged with 200-fold-fewer organisms (106 CFU), there was no significant difference between the efficacy of tobramycin and other b-lactams. Thus, tobramycin is particularly valuable in treating more severe P. aeruginosa pneumonia, whereas a no. of different b-lactam agents are of equiv. value in less severe lung infections.

DOCUMENT TYPE: Journal CA Section: 1 (Pharmacology) An accurate, sensitive, and specific liq.-chromatog. method is described for measuring piperacillin (I) [61477-96-1] in plasma and urine. Plasma samples deproteinized with 2 vols. of MeCN contg. 1.2 mg of the internal std., p-nitrobenzene sulfonamide, per L are centrifuged. The clear supernate is evapd. under N, and the residue is reconstituted in 50 mL of the mobile phase (32/68 by vol MeCN/H2O, adjusted to pH 2.5 with 85% phosphoric acid), of which 10 mL is injected onto a reversed-phase (C-18) column. Urine samples are dild. 10-fold with distd. water, an equal vol. of MeCN contg. 3 mg of the internal std. per L is added, and 20 mL is chromatographed. Stability studies indicate that storage conditions are crit. for both plasma and urine. Piperacillin in plasma is stable at -70° for at least 6 wk, but 100% of it is degraded during the same time at -20°. Piperacillin in urine is also stable at -70° for 6 wk, but 20% is degraded during 6 wk at -20°.
