(10-6, 10-5, 10-4 mol/L) of the ECF-A tetrapeptides, Val-Gly-Ser-Glu (I) [61756-22-7] and Ala-Gly-Ser-Glu (II) [61756-28-3], and histamine [51-45-6] dose-dependently enhanced the nos. of human eosinophils, but not neutrophils, forming rosettes with complement-coated sheep red blood cells (EAC3b); there was no increase in the nos. of eosinophils or neutrophils forming rosettes with IgG-coated sheep red blood cells. Enhancement of EAC3b eosinophil rosettes by I, II, or histamine increased with incubation time; with I and II, most of the increase in rosette formation occurred within 40 min whereas with histamine, enhancement of rosette formation took place between 40 and 80 min. Thus ECF-A peptides and histamine attract eosinophils selectively in chemotaxis; they may also have a direct effect on the eosinophil membrane as shown by the apparent increase in the nos. of complement receptors.
