DOCUMENT TYPE: Journal CA Section: 1 (Pharmacology) The metab. of 14C-labeled glyceryl trinitrate (GTN) [55-63-0] was studied in male rats after single dermal application or repeated dermal application for 5 or 10 days. [14C]GTN and its metabolites (1-glyceryl mononitrate [624-43-1], 2-glyceryl mononitrate [620-12-2], 1,2-glyceryl dinitrate [621-65-8], and 1,3-glyceryl dinitrate [623-87-0] were detd. in the plasma, urine, topical skin and some tissues (the brain, heart, kidney and liver) by HPLC with synchronized accumulating radioisotope detector. There was a significant accumulation of radioactivity in some tissues after repeated dermal application, which resulted from the endogenous compds. biotransformed from [14C]GTN. [14C]GTN was not detected in the liver, kidney, or heart but detected in the brain (0.0017 nmol/g) after repeated application for 10 days. [14C]GTN and its metabolites were found in the topical area of skin, indicating that [14C]GTN was metabolized in the skin itself after application.
