Characterization of the self-assembled monolayer was confirmed by a reaction between amine terminal groups and thionylchloride of Rhodamine-B dye using fluorescence microscopy. Quant. 3-dimensional profiling was obtained for all modification processes by converting microscopic images to numerical values. The reaction steps were also analyzed using IR spectroscopy (FTIR). .

Estradiol is labeled by means of a 2-phase derivatization procedure with either dansyl chloride or laryl chloride. After derivatization the excess reagent is removed on a short amino-bonded column: the sulfonyl chloride functional group reacts with the amino groups causing removal of the label; the estradiol derivs. are not retained. Chromatog. of estradiol derivs. is performed with methanol-water mixts. as eluent. Chem. excitation is carried out by adding bis(2-nitrophenyl)oxalate and hydrogen peroxide dissolved in acetonitrile to the column eluate. Linearity is obsd. over at least 3 decades (10-6-10-9M) for both dansyl chloride and laryl chloride. A detection limit of 5 ′ 10-10M (50 fmol injected) has been achieved and the dansyl derivatization method has been applied to the detn. of estradiol in spiked serum. .
