DOCUMENT TYPE: Journal CA Section: 63 (Pharmaceuticals) Microcryst. powder samples of DL-lysine acetylsalicylate (I) [62952-06-1], stored in vacuo at 25-70.degree., underwent autocatalytic degrdn. with 2nd-order rate kinetics; the kinetics were temp.-dependent in accordance with the Arrhenius law. In air, however, I decompn. followed the above kinetics only at 56-70.degree.; at 25-45.degree., more complex kinetics were obsd., probably due to adsorption of water on the I crystals, with subsequent hydrolysis, so that degrdn. was a function not only of I concn. but of the rate of hydrolysis in soln. and of the partial pressure of water. Calcns. from the above kinetics indicated that upon storage for 2 years at 25.degree. in vacuo and in air, the content of free salicylic acid in I would be 1.1 and 3.1%, resp.

DOCUMENT TYPE: Journal CA Section: 1 (Pharmacology) In 4 healthy male subjects there was no difference in inhibition of collagen-induced platelet aggregation after the administration of 500 mg Aspisol (acetylsalicylic-lysine)(ASA) [62952-06-1] alone or at salicylate (SA) [69-72-7] steady-state (3 g SA daily). But the inhibition of tissue-ext.-induced platelet shape change was diminished and shortened by pretreatment with SA. Thus, SA does not inhibit the effects of ASA on human platelet aggregation in vivo in therapeutic dose ranges. The clin. importance of the SA/ASA-interaction on tissue-ext.-induced platelet shape change remains to be clarified.
