Urinary I excretion reached a max. at 12-24 h, and 48-h excretions in the urine and feces, resp., amounted to 2.7% and 86% of the dose.

Continuous treatment abolished this effect. This apparent tolerance could be attributed to the inhibitory action of chronic I treatment on UDP-glucuronyltransferase activity, but the mechanism of this inhibition was not elucidated. Liver microsomal protein concn., ethlymorphine N-demethylation, and BSP clearance were unchanged.
