DOCUMENT TYPE: Journal CA Section: 1 (Pharmacodynamics) The potential nephrotoxicity of 7 cephalosporins was investigated in rats. The antibiotics were administered in 9 sep. i.m. doses on 5 consecutive days. The parameters for assessing toxicity were the rate of excretion of renal ephithelial cells, and the urinary enzymes malate dehydrogenase, lactate dehydrogenase, and glutamic-oxalacetic transaminase, which were detd. before, during and after treatment in 12-h urine voided overnight. In addn., serum urea was detd., and histol. examn. of the kidneys was performed. The following toxic threshold doses for the individual substances were derived from the analyses on the basis of cell excretion, the most sensitive parameter: cephaloridine [50-59-9] 500 mg/kg/day, cephradine [38821-53-3] 1000 mg/kg/day, cefazolin [25953-19-9] 2000 mg/kg/day, EMD 30087 [63521-15-3] 2000 mg/kg/day, cephacetrile [23239-41-0] 3000 mg/kg/day, cephalothin [153-61-7] 3000 mg/kg/day, cephapirin [21593-23-7] 5000 mg/kg/day. This type of investigation therefore permits cephalosporin antibiotics to be arranged in order of renal tolerance.
