Controlling primary hepatocyte adhesion and spreading on protein-free polyelectrolyte multilayer films
Controlling primary hepatocyte adhesion and spreading on protein-free polyelectrolyte multilayer films. Kidambi, Srivatsan; Lee, Ilsoon; Chan, Christina (Department of Chemical Engineering and Material Science, Michigan State University, East Lansing, MI 48824, USA). Journal of the American Chemical Society, 126(50), 16286-16287 (English) 2004 American Chemical Society. CODEN: JACSAT. ISSN: 0002-7863. DOCUMENT TYPE: Journal CA Section: 63 (Pharmaceuticals) Section cross-reference(s): 36 The development of new methods for fabricating thin films that provide precise control of the three-dimensional topog. and cell adhesion could lead to significant advances in the fields of tissue engineering and biosensors. This communication describes the successful attachment and spreading of primary hepatocytes on polyelectrolyte multilayer (PEM) films without the use of adhesive proteins such as collagen or fibronectin. We demonstrate that the attachment and spreading of primary hepatocytes can be controlled using this layer-by-layer deposition of ionic polymers. In our study, we used synthetic polymers, namely poly(diallyldimethylammonium chloride) (PDAC) and sulfonated poly(styrene) (SPS) as the polycation and polyanion, resp., to build the multilayers. 9003-53-6 and 63589-56-0 are also in the experiment. Primary hepatocytes attached and spread preferentially on SPS surfaces over PDAC surfaces. SPS patterns were formed on PEM surfaces, either by microcontact printing of SPS onto PDAC surfaces or vice versa, to obtain patterns of primary hepatocytes. PEM is a useful technique for fabricating controlled co-cultures with specified cell-cell and cell-surface interactions on a protein-free environment, thus providing flexibility in designing cell-sp. surfaces for tissue engineering applications. .
Material and method using polyanions to reduce nonspecific binding of a labeled material
Material and method using polyanions to reduce nonspecific binding of a labeled material. Hilerio, Fred J.; Kirchanski, Stefan J. (Becton Dickinson and Co., USA). U.S. US 5098849 A 24 Mar 1992, 9 pp. (United States of America). CODEN: USXXAM. CLASS: ICM: G01N033-566. ICS: G01N033-53; C12Q001-00. NCL: 436501000. APPLICATION: US 88-218712 13 Jul 1988. DOCUMENT TYPE: Patent CA Section: 9 (Biochemical Methods) A material and a method are described which use polyanions to reduce the nonspecific binding of a labeled material comprising a fluorochrome and a specific binding pair member. Also disclosed is an erythrocyte lysing soln. that reduces the nonspecific binding of a labeled material, the soln. 63589-56-0 and 68-04-2 are also in the experiment. comprising a C1-4 aliph. aldehyde, an alkali or alk. earth salt of a weak acid, a polyhydric alc., and 31 polyanion. The labeled material is esp. used to identify and detect cell populations within a sample of blood. The nonspecific binding of anti-Leu M3 monoclonal antibody (to CD14 antigen) conjugated with phycoerythrin was reduced in a blood sample by including Na heparin in an erythrocyte lysing soln. contg. formaldehyde, Na citrate, and diethylene glycol. .
