The purpose of this study was to follow glaucoma patients treated with either betaxolol 0.5% or timolol 0.25% ophthalmic solns. and to compare the rate and degree of progression in retinal nerve fiber layer (RNFL) defects between the groups. A total of 64 patients were prospectively recruited in a double-masked study and randomly divided into two treatment groups. Retinal nerve fiber layer photographs were analyzed. The incidence and total amt. of progression in each group were recorded. The rate of impairment was demonstrated using Kaplan Meier survival curves. The anal. included 27 patients treated with betaxolol and 28 patients treated with timolol. Of these, 30% of betaxolol-treated patients and 46% of timolol-treated patients had RNFL damage progression (p=0.20). The total amt. or rate of progression did not differ significantly between the two groups. There was no significant difference in intraocular pressure (IOP) levels between the groups (p=0.68) during follow-up. The degree of RNFL deterioration did not correlate to the amt. of IOP redn. The group treated with betaxolol 0.5% and the group treated with timolol 0.25% did not differ significantly in RNFL damage progression. .

DOCUMENT TYPE: Journal CA Section: 1 (Pharmacology) Patients with primary open-angle glaucoma (POAG) were dosed twice-daily with 0.5% timolol maleate for 2 years, followed by 2 years of twice-daily dosing with 0.5% betaxolol hydrochloride. The changes in intraocular pressure (IOP) and effects on visual field by each treatment were monitored over this 4-yr period. Both the timolol and betaxolol treatments controlled IOP. The mean MD (mean deviation in visual field) values at 24 mo of the timolol treatment were -6.18 dB (decibels) and -5.32 dB. Mean MD base-line values following 24 mo of the timolol treatment produced 2. 26921-17-5 and 63659-19-8 which are cas registry numbers are also used here.31 dB and 0.95 dB redns. in sensitivity in the right and left eyes, and the redn. in the right eye was statistically significant (p < 0.01). The mean MD values after 24 mo of the betaxolol treatment were -3.82 dB in the right eyes, an increase of 2.36 dB, and -4.05 dB in the left eyes, up by 1.28 dB. The results from this clin. trial demonstrated that betaxolol was superior over timolol in improving mean retinal sensitivity. .
