DOCUMENT TYPE: Journal CA Section: 16 (Fermentation and Bioindustrial Chemistry) For the sepn. and detn. of the closely related, cyclic undecapeptides cyclosporin A [59865-13-3], B [63775-95-1], C [59787-61-0], and D [63775-96-2] in samples from fermns., a high-speed liq. chromatog. method with conventional app. has been developed, which is based on a sepn. with a 3-mM Nucleosil C8 column and acetonitrile-water-phosphoric acid (:0.1) as eluent at 70°. The anal. time of ~3.5 min, the use of a sampler for 195 vials, and a lab. data system offer a high degree of automation and anal. economy. The coeff. of variation is ~0.8-1.5%. The rather low plate counts for the cyclosporins are due to the structures and high mol. wts. of these substances.

(from 25 to 75°) for cyclosporin A (I) [59865-13-3] and cyclosporin D (II) [63775-96-2]. The peak heights and no. of theor. plates (N) increased. The height equiv. to a theor. plate (HETP) decreased. The areas under the peaks, retention times and capacity factors (k') for both compds. did not vary with temp. With increasing eluent flow rate (from 0.5 to 2.5 mL/min), the peak heights, peak areas, retention times and N all decreased for both compds. A slight decrease in the capacity factor in I and II was also obsd. HETP increased with increasing flow. The sepn. factor, a, remained relatively const. for the ranges of temps. and flow rates investigated.
