DOCUMENT TYPE: Journal CA Section: 2 (Mammalian Hormones) Two RIAs, the 1st specific for 6-oxo-PGF1a [58962-34-8] and the 2nd for 6-oxo-PGF1a, 13,14-dihydro-6-oxo-PGF1a [71310-60-6], 13,14-dihydro-6,15-dioxo-PGF1a [63983-53-9], and 6-oxo-PGE1 [67786-53-2] were described. These RIAs were used to measure levels of immunoreactive 6-oxo-PGF1a and alleged metabolites of prostacyclin in human venous plasma. Procedures for the direct measurement and extn. of plasma 6-oxo-PGF1a were described and the limitations to which RIA of plasma 6-oxo-PGF1a is exposed were discussed. Direct measurement of plasma immunoreactive 6-oxo-PGF1a gave maximal levels of 6.1 pg/mL and 4 pg/mL in the 1st and 2nd RIAs, resp. The latter value reflected combined levels of <2 pg/mL of 6-oxo-PGF1a and cross-reacting metabolites in venous blood. Extn. of human plasma gave 88.7% recovery of [3H]6-oxo-PGF1a and 80-86% recovery of exogenous 6-oxo-PGF1a as immunoreactive 6-oxo-PGF1a. Basal plasma levels of extractable immunoreactive 6-oxo-PGF1a were <2.5 pg/mL. Prostacyclin [35121-78-9] incubated in vitro with blood was recovered in plasma as immunoreactive 6-oxo-PGF1a and confirmed that conversion to metabolites that cross-reacted with the 2nd antibody, in particular 6-oxo-PGF1, did not occur under exptl. 35121-78-9 and 58962-34-8 which are cas registry numbers of chemicals are mentioned. conditions. Extn. of [3H]6-oxo-PGF1a from acidified plasma with MeOH resulted in formation of a prostanoid that had properties consistent with the methylated hemiketal isomer of 6-oxo-PGF1a. Under RIA conditions this prostanoid was less immunogenic than native [3H]6-oxo-PGF1a and [3H]6-oxo-PGF1a extd. from plasma using Me formate. The low levels of plasma 6-oxo-PGF1a reported questions the validity of clin. studies that previously described altered levels of plasma 6-oxo-PGF1a in pathophysiol. conditions. .
