DOCUMENT TYPE: Journal CA Section: 1 (Pharmacodynamics) Colchicine [64-86-8] and vincristine sulfate [2068-78-2] differentiated sharply between hepatocyte mitosis assocd. with th Ehrlich ascites tumor, and regenerative (post-CCl4 necrosis) hepatocyte mitosis. In the former case there was a gross depression of prophase index and a marked lowering of the overall mitotoc index. In the latter, although a degree of prophase depression was evident, this was insufficient to prevent an overall substantial elevation of mitotic index due to accumulation at metaphase. Very low dose of colchicine claimed to be effective in stathmokinesis of hepatocytes under influence of hepatomitogenic tumor brei and tumour exts., produced no discernible effect on the mitotic pattern of hepatocytes in Ehrlich ascites tumor-bearing mice nor in post-CCl4 regenerating livers. The results are discussed in the context of known features of tumor-assocd. hepatocellular mitosis.

DOCUMENT TYPE: Journal CA Section: 1 (Pharmacodynamics) Chlorpromazine-HCl [69-09-0] (3×10-5 M) dibucaine-HCl [61-12-1] (10-5 M), lidocaine [137-58-6] (10-3 M) and procaine-HCl [51-05-8] (5×10-5 M) inhibited both the synthesis and secretion of plasma protein, but did not affect the uptake of L-leucine into rat liver slices nor the incorporation of phosphate into intracellular nucleotide phosphates or into phospholipids. The inhibition of secretion elicited by these drugs is probably not due to the inhibition of protein synthesis since cycloheximide at a concn. which completely inhibited protein synthesis, did not inhibit plasma protein secretion, while cycloheximide plus procaine did inhibit secretion and also caused a retention of nonsecreted plasma proteins within the slices. Unlike colchicine [64-86-8], however, procaine did not cause the retained plasma proteins to accumulate in Golgi-derived secretory vesicles, but showed a more general effect causing a distribution among several cell fractions.
