DOCUMENT TYPE: Journal CA Section: 1 (Pharmacology) Animal expts. indicated that vecuronium (I) [50700-72-6] and atracurium [64228-79-1] are short to medium duration neuromuscular blocking drugs with little tendency to cumulate and with a rapid rate of recovery, whereas pipecuronium [68399-58-6] is a long duration agent with a slow recovery rate and pronounced cumulative propensity. All 3 of these new agents are essentially free from cardiovascular side-effects. Their place in anesthetic practice will be detd. by clin. experience of their use, and by their different pharmacokinetic behavior including distribution, metab. and excretion, which are only meaningfully measured in man. Pancuronium [15500-66-0], the established drug, has an intermediate duration of action between those of vecuronium and atracurium on the one hand and pipecuronium on the other. Pancuronium has a slight tendency to produce increases in heart rate and blood pressure, effects which may be desirable under certain circumstances.

Octadecyl C-18 extn. columns were used for sepn. of the drugs from the plasma. Total chromatog. time between successive samples is 14 min. Recovery of atracurium is about 85%. Linearity over a concn. range of 10-2000 ng/mL is r2 = 0.998. The detection limit of the assay is 2 ng/mL. This method is reliable and sensitive enough for pharmacokinetic use in infants and children.
