DOCUMENT TYPE: Report CA Section: 4 (Toxicology) Single dose toxicities of 6 hexamethylmelamine [645-05-6] samples with different particle size distributions were evaluated in rats using the intragastric route. The LD50 values for the 6 samples were in the range 10,237 to 12,899 mg/m2. The sample with smaller particles (39 m) and the most uniform size distribution was more toxic than the sample contg. larger particles (95 m). Severity of toxic effects, however, was not strictly correlated with particle size since a sample with 35 m particles was less toxic than samples with particles in the 61-77 m range. Dose level had a significant effect on the quant. depression of blood leukocytes, lymphocytes, platelets, and body wt. in all samples. Particle size appeared to have minimal effect on these exptl. variables. The drug produced toxic effects on rapidly proliferating tissue; lymphoid, hematopoietic and germinal epithelium. A marked toxic effect on erythrocytes was evident as anemia or hemoconcn. and elevated myeloid-erythroid ratios in bone marrow. Histologic evidence of cellular depletion included lymphocytolysis and hypoplasia in lymphoid tissue, hypoplastic bone marrow, and spermatogenic arrest in germinal epithelium. The major target organs at nonlethal doses were the bone marrow and germ cells, with germinal epithelium showing the most severe lesions.

Non-alkylating chemosterilants, hempa [680-31-9] and hemel [645-05-6], did not induce any change in the ovary even at high doses although they produced sterility in the female.
