DOCUMENT TYPE: Conference CA Section: 1 (Pharmacodynamics) Section cross-reference(s): 8, 2 In patients with various indications for vitamin D therapy or normal subjects who received long term treatment with vitamin D2 [50-14-6] or vitamin D3 [67-97-0] (at least 4 mo), there was a clear relation between vitamin D dosage and blood plasma 25-hydroxyvitamin D (I) [64719-49-9] levels. In subjects receiving 25-hydroxycholecalciferol (25-OHD3) [19356-17-3] (for at least 6 weeks) the relation between dosage and plasma I levels was less obvious. Within the dose range studied, 25-OHD3 was 8-13 times more potent than vitamin D on a wt. basis in producing equiv. plasma levels of I. In life guards in midsummer, plasma I was equiv. to that in patients receiving 8,000-10,000 IU vitamin D daily. Short term treatment with UV-light produced plasma I levels equiv. to 8,000-10,000 IU vitamin D. Again, 25-OHD3 was 10 times more potent than vitamin D. There appears to be constraints on the 25-hydroxylation of vitamin D, with only approx. 1/10 of the administered dose of vitamin D relative to 25-OHD3 appearing as plasma I.
