DOCUMENT TYPE: Journal CA Section: 1 (Pharmacodynamics) Urapidil-HCl (I) [64887-14-5] was screened in mice, rats, guinea pigs, and cats in a battery of tests relating to sportaneous behavior, reactive behavior, and autonomic and organ functions. I had a central sedative action in mice and rats at oral doses >10 mg/kg; for doses up to ~100 mg/kg, this sedation could be interrupted by external stimuli, and in this respect I differs from other centrally acting drugs such as chlorpromazine, diazepam, and clonidine. I seemed not to block central dopamine receptors, since it inhibited neither the conditioned avoidance reflex nor amphetamine-induced gnawing in mice and rats. I differed from tranquilizers such as diazepam in that it did not inhibit the spinal polysynaptic motor reflexes of the decerebrate cat. At <80 mg I/kg, orally, there were no hypotensive effects in rats, so that the obsd. changes in behavior were not due to acute changes in cardiovascular function. The antihistamine effect of I on the isolated guinea pig ileum was ~7% that of chlorpromazine. Further I had no anticholinergic activity.

The calibration curve of urapidil hydrochloride had good linearity within the range of 3.2-11.2 mg/L (r = 0.9998). The av. recovery of urapidil hydrochloride was 99.5% with the relative std. deviation (RSD) of 1.7%, and the av. within-day precision and day-to-day precision were 1.3% and 2.2%, resp. There was no statistical difference in the content of urapidil hydrochloride during i.v. injection via micro-pump for 9 h. The results showed that there was no adsorption of urapidil hydrochloride by syringe during i.v. injection via micro-pump. .
