DOCUMENT TYPE: Journal CA Section: 64 (Pharmaceutical Analysis) The Japanese Pharmacopoeia (JP) serves as the ref. std. for the quality of medicinal drugs. Identification methods based on comparison of spectral patterns of samples with ref. spectra have been introduced in many monographs in JP14, using not only IR (IR) spectra, but also UV and Visible (UV) spectra. There are some problems in judging identities by means of these methods. Therefore, in this paper, the authors consider the problems and clarify their causes, and also offer advice for carrying out the test precisely. For example, anion-exchange may occur when salt-type drugs, such as croconazole hydrochloride, penbutolol sulfate and oxapium iodide, are mixed and ground with potassium bromide (KBr) in the course of prepg. tablets for the measurement of their IR spectra by the KBr disk method. To avoid the spectral change due to such anion-exchange, it is desirable to use the potassium chloride (KCl) disk method to measure the IR spectra of drugs of chloride salt-type, and to use the paste method with liq. paraffin to measure the IR spectra of drugs of other salt-types.In this experiment, several chemicals are used like 6577-41-9 and 38363-32-5 Therefore, IR spectra measured by the KCl disk method were adopted as ref. spectra for 34 kinds of drugs of chloride type in JP14. .
