DOCUMENT TYPE: Journal CA Section: 2 (Mammalian Hormones) delta sleep-inducing peptide (DSIP) [69431-45-4] or 5 closely related peptides were injected peripherally, and the levels of DSIP-like immunoreactivity were measured in the brains of 100 g and 500 g rats decapitated a minute later after washout with 0.9% NaCl. Higher concns. of immunoreactive material were found in brain tissue with injection of 1-de-tryptophan-DSIP [62568-58-5], 4-D-alanine-DSIP [65908-44-3], and, after correction for cross-reactivity, 4-D-alanine-DSIP-NH2 [84609-38-1] than with the other peptides. Both peptides substituted in the 4th position with D-alanine resulted in higher concns. in 500 g rats than in 100 g rats, indicating a possible effect of age. The 5-10 fold greater brain to blood ratios of radioactivity after 125I-labeled N-tyrosine-DSIP [65908-45-4] than after 125I radioiodinated serum albumin provided addnl. evidence against nonspecific leakage or contamination of the brains with substantial amts. of residual blood. Penetration of the blood-brain barrier by small amts. of peptides, at least DSIP peptides, may be somewhat selective. .
