In dispersed acini from mouse or rat pancreas, I stimulated amylase secretion. The stimulation of amylase secretion caused by a maximally ED of I was less than that caused by a maximally ED of II, and in contrast to the action of II supramaximal concns. of I did not cause submaximal stimulation of amylase secretion. Dibutyryl cGMP [32266-35-6] and proglumide [6620-60-6] each inhibited the stimulation of amylase secretion caused by I. I inhibited binding of 125I-labeled cholecystokinin [9011-97-6] to mouse and rat pancreatic acini. Thus, in mouse and rat pancreatic acini, I is a partial agonist and this partial agonist activity is produced by occupation of the cholecystokinin receptor by I.

Water intake also increased 18 h after treatment and there were no differences between the lean and obese rats. Feeding patterns in 6-h fasted obese rats treated with 0.59 g proglumide were characterized by increased meal frequency but smaller meal size. Proglumide probably increased feeding in the Zucker rats by antagonizing the cholecystokinin [9011-97-6] receptor regulating satiety.
