For the synthesis of ribozyme regions with phosphorothioate linkages, a 0.05 M soln. of 3H-1,2-benzodithiol-3-one 1,1-dioxide in acetonitrile was used in the reaction (Wincott, F., et al, 1995; Usman, N., et al, 1987). The automated sulfurization of ribose and other 2'-substituted nucleotides in the context of oligonucleotide synthesis was optimized. Anion exchange chromatog. was used to purify the synthesized ribozyme. The purified ribozyme showed catalytic activity. .
