DOCUMENT TYPE: Journal CA Section: 2 (Mammalian Hormones) A method of examg. the catabolism of neuropeptides in the tissue fluids that bathe specific regions in the alimentary tract of the conscious animal is described. Two bundles of dialysis fibers were surgically implanted in the submucosal region of the gastric corpus in the anesthetized rat. After recovery from surgery, one set of fibers was perfused with 125I-labeled Tyr4-bombesin 14 [67338-70-9]. Some mols. of peptide diffused into the tissues and were catabolized by enzymes therein. The catabolites were collected in the 2nd dialysis fiber and fractionated by HPLC. Bombesin 14 was catabolized to iodo-tyrosine and 3 unidentified peptides. The catabolites did not bind with a carboxyl-terminal specific antiserum and, as this is the biol. active region of bombesin, were devoid of any biol. activity.

DOCUMENT TYPE: Journal CA Section: 2 (Mammalian Hormones) The binding of 125I-labeled 4-tyrosine-bombesin (125I-Tyr4-bombesin) [67338-70-9] to rat brain slices was investigated. Radiolabeled bombesin bound with high affinity (dissocn. const. = 4 nM) to a single class of sites (max. binding capacity = 130 fmol/mg protein); the ratio of specific to nonspecific binding was 6/1. Also, pharmacol. studies indicated that the C-terminal of bombesin was important for the high-affinity binding activity. Autoradiog. studies indicated that the radiolabeled bombesin-binding sites were discretely distributed in certain gray but not white matter regions of rat brain. Highest grain densities were present in the olfactory bulb and tubercle, nucleus accumbens, suprachiasmatic and periventricular nuclei of the hypothalamus, central medial thalamic nucleus, medial amygdaloid nucleus, hippocampus, dentate gyrus, subiculum, nucleus of the solitary tract, and substantia gelatinosa. Moderate grain densities were present in the parietal cortex, deep layers of the neocortex, rhinal cortex, caudate putamen, stria terminalis, locus ceruleus, parabrachial nucleus, and facial nucleus. Low grain densities were present in the globus pallidus, lateral thalamus, and midbrain. Negligible grain densities were present in the cerebellum, corpus callosum, and all regions treated with 1 mM unlabeled bombesin. The discrete regional distribution of binding suggests that endogenous bombesin-like peptides may function as important regulatory agents in certain brain loci.
