DOCUMENT TYPE: Journal CA Section: 1 (Pharmacology) Section cross-reference(s): 11 A 95% ethanolic crude ext. of Shan-Dou-Gen (root of S. subprostrata) yielded several components such as b-sitosterol [83-46-5], stigmasterol [83-48-7], lupeol [545-47-1], b-sitosterol-b-D-glucoside [474-58-8], 1-maackiain [2035-15-6], sophoranone [23057-55-8], sophocarpine [6483-15-4], trifolirhizin [6807-83-6], and matrine [519-02-8]. The effects of i.p. administration of each fraction, subfraction, or pure compd. obtained from Shan-Dou-Gen on the thermoregulatory responses (including metabolic rate, vasomotor activity and rectal temp.) were assessed in conscious rats at various ambient temps. (Ta). Injection of matrine or crude ext. of Shan-Dou-Gen produced hypothermia in rats at Ta of 8° and 22°. The hypothermia in response to matrine was due to a decrease in metabolic rate and (or) an increase in cutaneous temp. However, i.p. injection of matrine or crude ext. of Shan-Dou-Gen produced an insignificant change in thermoregulatory responses in rats at Ta of 30°. In addn., the hypothermic effect of matrine was not antagonized by pretreatment of rats with 5,6-dihydroxytryptamine or 6-hydroxydopamine. Apparently, matrine is the active hypothermic constituent of the root of S. subprostrata; the hypothermic effect of matrine appears not to be assocd. with the central release of serotonin or catecholamines.
