The aim of the present expts. was to evaluate if ischemia and reperfusion modify histamine and lactate dehydrogenase (LDH) release in isolated guinea pig heart. A steady increase of LDH release was obsd. both in the ischemic and reperfusion phases. The release of histamine was reduced during the ischemic phase and increased during reperfusion. A diminution of mast cell granule metachromasia was obsd. in the right auricles at the end of the reperfusion period. D-Mannitol and reduced glutathione modified the kinetics of histamine and LDH release. Cimetidine was able to decrease the release of histamine during the ischemic and reperfusion phases and also reduced the release of LDH; triprolidine was completely ineffective. Apparently, O-derived free radicals may be involved in the pathogenesis of myocardial dysfunction after ischemia and reperfusion. .

The saponins were detected by general test for saponins and pos. tests for a triterpenoid. The sugars obtained on hydrolysis of saponins were identified as D-glucose and D-galactose, the latter being present in traces. The triterpenoid sapogenins were found to be a mixt. of 2 compds. which have been isolated. The phys. and the chem. data support the identities of the compds. as randialic acids A and B. The nonsaponin fraction was isolated through the lead salt. The lead acetate complex on treatment with H2S yielded scopletin, which was methylated to provide 6,7-dimethoxycoumarin (scoparone).
