Consistent with inhibition of DA synthesis, administration of both a-MT and MIT resulted in marked redns. in the hypothalamic concns. of DA and its metabolite homovanillic acid [306-08-1] as well as in increases in prolactin [9002-62-4] secretion. a-MT administration, but not MIT, resulted in a decrease in NA concn. and a increase in the concn. of the NA metabolite 3,4-dihydroxyphenylethyleneglycol (DHPG) [3343-19-9]. The hypothalamic DHPG/NA ratio was thus markedly increased by a-MT indicating increased NA neuronal activity. a-MT administration also increased ACTH [9002-60-2] secretion, an effect not obsd. following MIT. Apparently, the effects on hypothalamic NA activity and ACTH secretion caused by a-MT are stress-mediated and unrelated to tyrosine hydroxylase inhibition. MIT is devoid of these effects but exhibits DA synthesis blockade activity, thus indicating it to be a preferable drug for the acute inhibition of tyrosine hydroxylase in neuroendocrine investigations.

DOCUMENT TYPE: Journal CA Section: 1 (Pharmacology) Section cross-reference(s): 2 In mice given 3-day i.v. infusions of large doses (15 or 30 mM, 1.8 mL/day) of mono-iodotyrosine [70-78-0] and di-iodotyrosine [300-39-0], some of the mammary tumor (MT)- and adrenocortical tumor (AT)- grafts transplanted 9 or 15 wk after infusion disappeared, whereas those transplanted 3 wk post-infusion remained unaffected. MT grafts also disappeared in some thyroxine [51-48-9]-infused hosts, although the thyroxine infusion had no suppressive effects on AT grafts. I.V. infusion of NaI and tyrosine have no suppressive effect on MT and AT grafts suggesting that iodine and tyrosine presumably derived from iodotyrosines exert no inhibitory effect on the tumor. .
