DOCUMENT TYPE: Journal CA Section: 63 (Pharmaceuticals) Copoly(amino acids) having both hydrophilic and hydrophobic residues, copoly(g-benzyl L-glutamate-N5-dihydroxyethylaminopropyl-L-glutamine) (I) [105681-92-3], was prepd. by polymn. and cast to a 20-50 mm thick film. A sponge sheet prepd. by freeze drying of 100 mL aq. soln. of modified collagen (1 g) and pepleomycin sulfate (II) [70384-29-1] (1 g) was pressed to give a disk. A disk laminate was prepd. by covering with 2 film layers. Drug permeability through the films contg. 30 mol. % hydrophilic residue were in the order: 5-fluorouracil [51-21-8] > Na prednisolone 21-succinate [1715-33-9] > adriamycin [25316-40-9] > II. The drug permeability increased to very high levels when more hydrophilic film was used, whereas it was too low when less hydrophilic film was used. A drug releasable laminate under control contg. 10 mg II was implanted s.c. in rat femur, and it was found that no capsule formation was obsd. around the laminate implanted. The I film of 35 mm thick contg. 30 mol. % hydrophilic residue was the best material for controlled release of II.
