DOCUMENT TYPE: Conference CA Section: 1 (Pharmacology) To evaluate the immune cellular effect of pefloxacin (PEF)(I) [70458-92-3], the mitogen proliferative responses of human peripheral blood mononuclear leukocytes (PBL) at different antibiotic concns. were studied. The response to PHA at optimal concns., as measured by incorporation of [3H]Tdr, was not modified at therapeutic doses (0.1-10 mg/mL) of PEF. The inhibition of the proliferative response was obtained only with high antibiotic concns. (>25 mg/mL). Similar results were obtained with the 2 main metabolites of PEF. There did not appear to be an interaction of O exposure with the PEF effect. Two other quinoline derivs. a [ciprofloxacin [85721-33-1] and ofloxacin [82419-36-1]] had the same effect as PEF. .

DOCUMENT TYPE: Journal CA Section: 1 (Pharmacology) Serum protein binding, extravascular diffusion, and urinary excretion of pefloxacin (I) [70458-92-3] were studied in rabbits; the effect of furosemide [54-31-9] on the urinary excretion of pefloxacin was also investigated. Serum protein binding of the drug was 25%; extravascular concns. were within the range of the minimal inhibitory concns. for most susceptible bacterial strains. Pefloxacin appeared to be reabsorbed by renal tubules. Furosemide increased the renal excretion of pefloxacin through a tubular process. Penetration of pefloxacin into normal heart valves and Escherichia coli-infected vegetations was good; pefloxacin reduced the colony counts in the infected vegetations after 7 i.m. injections.
