DOCUMENT TYPE: Journal CA Section: 26 (Biomolecules and Their Synthetic Analogs) The paper illustrates two efficient routes to macrolactone I contg. a 3-(para-methoxybenzyloxy)-Pr side chain at C-15. The chiral center at C-15 was introduced by a Noyori redn. of keto ester (II). The intermediate common to both routes, aldehyde III, was prepd. from II. The subsequent chain extension utilized Evans aldol reactions. The first route leads to the alkene, which was used, after hydroboration, for a Suzuki cross-coupling reaction with vinyl iodide. The derived seco acid was converted into the macrolactone I by a Mitsunobu lactonization by using immobilized triphenylphosphine. Alternatively, an aldol reaction of aldehyde III with the 4-pentenoyl deriv. was used to prep. alkene. This building block led to ester, which could also be converted into I by the classical ring-closing metathesis. After conversion of the C-15 side chain to the corresponding aldehyde, the enamide was introduced through hemiaminal formation and formal elimination of water. Sepn. 72287-26-4 and 825612-59-7 are just another two chemicals used in this study. of the double-bond isomers and removal of the silyl protecting groups provided salicylihalamides A (E)-II and B (Z)-II. .
