Ca gluceptate (CaG) [29039-00-7], FeSO4.H2O, metoclopramide-HCl (MCP) [7232-21-5], methenamine (MTA) [100-97-0], and sucrose (S) [57-50-1] prepd. on a single-punch tablet press using both unlubricated and lubricated dies. The materials were directly compressible and included solids which exhibit plastic deformation and brittle fracture on compression. Surface recrystn. and changes in the matrix of the compact were obsd. after storage at 25° and 0 and 43% relative humidity. Crystal growth rates were more obvious for ASA, CaG and MCP, where extensive surface recrystn. was obsd. within 60 min of compression. Recrystn. of MCP and MTA led to complete reorganization of the surface. Unlike the other solids, the surface of MTA compacts depended on lubrication. The surface of a lubricated compact was initially smooth, but without lubricant, it was highly disordered. After about 8 days and considerable structural reorganization, no significant difference remained between MTA compacts prepd. with or without lubrication. Recrystn. occurred much more slowly with S and FeSO4.H2O, but surface growth and restructuring of the surface were apparent after about 12 wk. Examn. of diametrally fractured compacts showed recrystn. at crystal interfaces within the compact. It is suggested that the activity of some solids increases on compaction, possibly through the creation of lattice defects at the crystal surfaces. The consequent decrease in free energy necessary to restore thermodn. equil. could explain the obsd. crystal growth, and may be responsible for bonding between the particles.

On the other hand, administration of sulpiride [15676-16-1] (1.5 mg/kg) and Paspertin (metoclopramide) [7232-21-5] (0.15 mg/kg) i.v. had no effect on cerebral or pituitary circulation whereas infusion of dopamine [51-61-6] (60 mg/kg/min, i.v.) and injection of bromocriptine [25614-03-3] (75 mg/kg) slightly decreased pituitary local flow. Results are compared to those using other techniques and the action mechanism of bromocriptine in decreasing pituitary tumors.
