DOCUMENT TYPE: Journal CA Section: 6 (General Biochemistry) Section cross-reference(s): 14 When purified normal human Hb was incubated with labeled glucose, a labeled product was produced which had properties identical to those of Hb AIc, and the glucose was shown to be added at the N terminus. This was independent of the N-terminal sequence and was apparently a nonenzymic reaction. The structure of the N-terminal product most consistent with the chem. properties of Hb AIc (e.g., NaBH4 redn. and spectra) was that of an N-glycoside in equil. with a Schiff base. Expts. in which D-glucose-2-3H or D-glucose-5-3H were incubated with erythrocytes showed the intracellular occurrence of an Amadori rearrangement of the initial glucosylated Hb product (Schiff base form) to produce a 1-amino-1-deoxy-2-keto deriv. of glucose with a 5-membered ring. The significance of this nonenzymic Hb glucosylation for diabetes, in which higher than normal Hb AIc concns. are present, is indicated.Except for chemicals metioned above, 9034-51-9 and 7284-37-9 are also used. .
