DOCUMENT TYPE: Patent CA Section: 16 (Fermentation and Bioindustrial Chemistry) L-Threo-b-hydroxyaspartic acid (I) [7298-99-9] was prepd. by fermn. with Dactylosporangium. Thus, a seed culture of Dactylosporangium SF-2253 (FERM-P6858) was inoculated into 35 L medium (pH 7) contg. malt syrup 2, soybean oil 0.15, soybean flour 1, distillers' spent grain 0.25, cottonseed oil 0.5, CaCO3 0.1%, and traces of FeSO4, NiCl2, and CoCl2 and incubated at 28° for 5 days with stirring and aeration. The broth filtrate (40 L) was brought to pH 2 and passed through a silica gel column. I was adsorbed to Dowex 1′2(Cl-) and eluted with 0.2M NaCl and the eluate was concd., filtered, and chromatographed on Sephadex G-10 to yield 31.5 g crude I, which was purified by chromatog. on cellulose and recrystn. from EtOH.
