DOCUMENT TYPE: Journal CA Section: 1 (Pharmacology) After oral administration of di-Me 4,4'-dimethoxy-5,6,5',6'-dimethylenedioxybiphenyl-2,2'-dicarboxylate (biphenyl di-Me dicarboxylate, BDD)(I) [73536-69-3], used for the treatment of chronic hepatitis, to rats a water-sol. metabolite was sepd. from urine by TLC. After hydrolysis of the metabolite with glucuronidase, a compd. was isolated by TLC and identified by mass, IR, and NMR spectra as di-Me 4-hydroxy-4'-methoxy-5,6,5',6'-dimethylenedioxybiphenyl-2,2'-dicarb oxylate (4-demethyl-BDD) [91828-88-5]. The actual water-sol. metabolite in urine is apparently 4-demethyl-BDD glucuronide [91828-89-6]. After oral administration of [3H]BDD to rats, the main part of the radioactivity in the liver and kidneys appears to be 4-demethyl-BDD glucuronide.
