DOCUMENT TYPE: Journal CA Section: 2 (Mammalian Hormones) The effect of motilin [52906-92-0] on vascular tone, cardiac rhythm, and blood vessel response to pressor stimuli was studied in the pithed rat. Motilin produces a prolonged depressor effect. The depressor responses were dose-dependent at 30-300 nmol/kg (max. decrease: -22 mmHg). In addn., motilin attenuated pressor responses to vasopressin [11000-17-2], leukotriene D4 [73836-78-9], and the pressor effect evoked by complete spinal cord stimulation. Motilin did not affect the basal heart rate nor did it alter sympathetically induced heart rate acceleration. Motilin did not affect the circulatory level of norepinephrine or epinephrine [51-43-4] at resting state or of norepinephrine released by spinal cord stimulation; motilin suppressed epinephrine released by spinal cord stimulation. Apparently, motilin has a role in regulation of blood vessel tone by direct action on the vascular smooth muscle. In addn., motilin might play a role in regulation of epinephrine release from the adrenal medulla.

After solvent extn. and purifn. on C18 Sep-Pak cartridges, tritiated products migrating on reversed-phase HPLC with authentic unlabeled LTC4 [72025-60-6], LTD4 [73836-78-9], and LTB4 [71160-24-2] were obsd. The identity of LTC4 was confirmed through enzymic conversion into LTD4 by g-glutamyl transpeptidase as well as by bioassay on the rat stomach fundus after HPLC purifn. The contractile response to the extd. material was blocked by the SRS antagonist, FPL 55712. LTB4 synthesis was located in the 100,000g supernatant, whereas LTC4 synthesis was present in the corresponding pellet. LTC4 formation was enhanced when reduced glutathione was supplemented in the incubation medium. These results demonstrate the presence in rat liver of various enzymic steps in LTA4 catabolism.
