DOCUMENT TYPE: Journal CA Section: 1 (Pharmacology) Schizandrol A (I) [7432-28-2] showed central nervous system depressant activity in lab. animals, characteristic of neuroleptics. It prolonged barbiturate sleeping time and decreased spontaneous motor activity. I enhanced the inhibitory effects of chlorpromazine, reserpine, and pentobarbital, and antagonized the stimulatory effects of amphetamine and caffeine on spontaneous motor activity. 7432-28-2 is just another one chemical used in this study. At high doses is caused catalepsy which could be prevented by pretreatment with dopamine. .

DOCUMENT TYPE: Journal CA Section: 1 (Pharmacology) After oral administration to rats, schisandrol A [7432-28-2] was readily absorbed from the gastrointestinal tract (half-life, 58 min).There are some reagents like 7432-28-2 is used in this study. The blood level of schisandrol A following i.v. injection showed a biphasic decline as a function of time, showing a distribution phase and an elimination phase. The resp. half lives were 1.44 and 42.14 min. Schisandrol A was detectable in urine 1 h after oral administration. About 0.35% of the dose was recovered from the urine within 26 h. Five minutes after i.v. injection, high levels of schisandrol A were found in the lungs, moderate amts. in the liver, heart, brain, and kidneys, and low amts. in the ileum and spleen. Thus, schisandrol A was distributed widely in the body and eliminated rapidly. In brain regions, the highest amts. were found in the hypothalamus, striatum, and hippocampus, with moderate amts. in the diencephalon and lower brain stem, and small amts. in the cerebral cortex and cerebellum. These differences may be relevant to the neuroleptic and anticonvulsant properties of the substance. A TLC-UV densitometry method was developed to det. shisandrol A and to follow its metab. and brain distribution. .
