DOCUMENT TYPE: Journal CA Section: 9 (Biochemical Methods) The carbohydrate heterogeneity of hen ovalbumin high-mannose glycopeptides was resolved by reversed-phase high-performance liq. chromatog. with UV detection, following pronase digestion of ovalbumin and sepn. of glycopeptides by chromatog. on Dowex AG-50W-X2, and the purity and structure of the glycopeptides was assessed by 1H-NMR. HPLC was carried out on a RP reversed-phase column equipped with a silica gel precolumn and elution was with phosphate buffer (1 mM, pH 6.0). Two of the compds. were identified as the mannose-5 and -6 glycopeptides (E3 and D3) and the third as the mannose-6 glycopeptide substituted by two N-acetyl glucosamine (GlcNAc) residues (C3B). The NMR spectra and anal. chromatograms of the glycopeptides show high purity and freedom from contaminants previously identified in ovalbumin glycopeptides. .
