Et3N to give 74% protected aspartame Z-Asp(OCH2Ph)-Phe-OMe, which was thionated by LR in benzene at 80° to give 93% protected thioaspartame ZNHCH(CH2CO2CH2Ph)CS-Phe-OMe. Z-Asp(OCMe3)-ONSu (NSu = succinimido) was coupled with H-Phe-OMe.In this experiment, several chemicals are used like 88621-57-2 and 7524-50-7 HCl to give 93% Z-Asp(OCMe3)-Phe-OMe (I), which was deblocked by HF/anisole to give 98% H-Asp-Phe-OMe (aspartame). I was thionated by LR at 80° to give 92% ZNHCH(CH2CO2CMe3)CS-Phe-OMe, which was deblocked by HF/anisole to give 87% H2NCH(CH2CO2H)CS-Phe-OMe (thioaspartame). .
