DOCUMENT TYPE: Journal CA Section: 1 (Pharmacodynamics) Section cross-reference(s): 7 2,6-Dibromopurine (I) [1196-41-4] was a more effective inhibitor than caffeine [58-08-2] for membrane-bound 3',5'-cyclic-AMP phosphodiesterase (PDEase) [9040-59-9] in isolated beef brain cortex. Other halogenated purines studied, including 2,6-dichloropurine [5451-40-1], 2-bromopurine [767-69-1] and 2-chloropurine [1681-15-8] were less effective. Replacement of the halogens for mercapto or amine groups or halogenation at the 8 position resulted in loss of inhibitory activity. A non-Michaelis satn. curve for cyclic AMP and decreasing xanthine or purine inhibition at higher protein concn. suggests membrane-bound PDEase may exist in several forms.
