DOCUMENT TYPE: Journal CA Section: 1 (Pharmacology) A liq. chromatog. screening procedure is described for the detection, in urine, of 12 of the 15 K+-depleting diuretics available in Australia. A 2-mL urine sample was acidified with NaH2PO4 (pH 4.1) and extd. with 4 mL Et acetate. The sample was cleaned up further by washing with 5 mL Na2HPO4 (pH 7.5). The Et acetate was then evapd. to dryness, the residue reconstituted in 100 mL mobile phase and 5 mL were injected onto a Merck LiChrosorb RP-18 (5 mm) column. The UV of the eluent was monitored at 271 nm for 10 min.In this experiment, several chemicals are used like 58-93-5 and 135-07-9 The screen was evaluated by giving each of 30 volunteers the lowest recommended dose of one of the diuretics in the study and obtaining urine samples 4, 8, and 24 h after having taken the dose. Twelve diuretics, chlorothiazide [58-94-6], hydrochlorothiazide [58-93-5], quinethazone [73-49-4], chlorthalidone [77-36-1], methyclothiazide [135-07-9], clopamide [636-54-4], frusemide [54-31-9], metolazone [17560-51-9], mefruside [7195-27-9], bendrofluazide [73-48-3], cyclopenthiazide [742-20-1], and bumetanide [28395-03-1], were all detectable up to 24 h after a dose. Thus, the screen would be reliable for the detection of these diuretics in urine. .

DOCUMENT TYPE: Journal CA Section: 1 (Pharmacology) Intragastric intubation of chlorthalidone [77-36-1] (3.0 mg/kg), hydrochlorthazide [58-93-5] (3.0 mg/kg), frusemide [54-31-9] (1.0 mg/kg) and clonidine [4205-90-7] (0.003 mg/kg) did not have an effect on fasting blood sugar level of rabbits up to 5 h. When clonidine was administered along with glibenclamide [10238-21-8] (0.024 mg/kg), it not only caused an early onset of hypoglycemia but also produced a greater fall in blood sugar than obsd. with glibenclamide alone. The diuretics did not modify the hypoglycemic activity of glibenclamide. .
