DOCUMENT TYPE: Conference CA Section: 13 (Mammalian Biochemistry) When exogenous H2O2 was introduced into rat liver cells at expected physiol. rates, metab. was principally regulated by glutathione peroxidase; at higher H2O2 infusion rates, metab. occurred mainly by catalase. However, when H2O2 was infused into suspensions of rat cardiac myocytes, no O evolution was detectable within the 1st minute of infusion, indicating that the cells are substantially free of catalase. H2O2 infusion caused rapid oxidn. of myoglobin to metmyoglobin, and concomitant with this oxidn., O evolution occurs. Consequently, the cardiac activity of heart cells appears to be largely due to metmyoglobin. Apparently, the glutathione peroxidase-glutathione reductase system has a principal role in H2O2 metab. under physiol. rates of formation in heart, and the overall efficiency of this system is greater in heart than in liver. .

DOCUMENT TYPE: Journal CA Section: 13 (Mammalian Biochemistry) Phenanthroline, a strong Fe chelator, prevents both the formation of DNA single-strand breaks and the killing of mouse cells produced by H2O2. These results, taken together with previous findings, indicate that the DNA damage is produced by hydroxyl radicals formed when H2O2 reacts with chromatin-bound Fe2+ and that this damage is responsible for the killing effect. .
