DOCUMENT TYPE: Journal CA Section: 2 (Mammalian Hormones) The pharmacol. characteristics and anatomical distribution of 3H-labeled a-amino-3-hydroxy-5-methyl-4-isoxazolepropionic acid (AMPA) [77521-29-0] binding sites in rat brain were examd. using quant. autoradiog. These binding sites exhibit the appropriate pharmacol. characteristics and are found in high concns. in the hippocampus, cerebral cortex (esp. layers I-III), induseum griseum, and dorsal lateral septum. Intermediate concns. are found in the corpus striatum and deeper layers of cerebral cortex. Lower concns. are found in the diencephalon, midbrain, and brainstem. Evidently, [3H]AMPA binding sites are found throughout the central nervous system and certain brain regions use quisqualate [52809-07-1] receptors as glutamate [56-86-0] neurotransmitter receptors.

DOCUMENT TYPE: Journal CA Section: 2 (Mammalian Hormones) The functional size of the quisqualate-type receptors was investigated by high-energy radiation inactivation anal. of the binding of 3H-labeled a-amino-3-hydroxy-5-methyl-4-isoxazolepropionic acid (AMPA) [77521-29-0] to rat cortical membranes. The AMPA binding site is coupled to a high-mol.-wt. modulatory unit, which down-regulates the affinity of the binding site for quisqualate [52809-07-1], glutamate [56-86-0], and AMPA. The estd. mol. target size of the AMPA binding site is different from the reported mol. wt. of the glutamate binding site isolated from bovine brain. Furthermore, enhanced [3H]AMPA binding induced by chaotropic ions seems not to be a consequence of removal of the modulatory unit.
