Mass spectral anal. and cochromatog. with authentic ref. compd. was utilized to identify the lactone ring-opened metabolite as o-hydroxyphenylethanol (II) [7768-28-7]. Formation of II required the microsomal and cytosolic fractions and the max. rate of prodn. was obsd. in the presence of NADPH and NADH. Anal. of urine from rats administered [14C]I established that II was formed in vivo and the amt. excreted was ~15% of that of o-hydroxyphenylacetic acid [614-75-5]. Thus, II was readily converted to o-hydroxyphenylacetic acid in vivo and in vitro while the reverse reaction was not obsd. The relations of these results to the currently accepted pathway(s) leading to ring-opened I products are discussed.
