P. fluorescens Is mutagenized0with UV light and grown in a medium contg. salts and 0.5% glucose (no inducer is added). The cells are harvested by centrifugation, disrupted in 0.02M phosphate buffer (pH 7.4), and centrifuged. The supernatant is the cell-free ext. contg. IDH. L-Sorbitol is oxidized in a reaction mixt. contg. 1 mmol L-sorbitol, 50 mL of 25 mM phosphate buffer (pH 8), 3 mmol NAD, methylene blue and cell-free ext. (20-50 mg protein). The mixt. is incubated at 30° for 2-3 h or until equil. is established, heated in a boiling water bath, and the denatured protein removed by centrifugation.
