DOCUMENT TYPE: Conference CA Section: 1 (Pharmacology) The metab. of doxazosin mesylate (I) [77883-43-3] was studied in man, rat, mouse, and dog using carbon-14 labeled drug. Bioavailability and pharmacokinetic studies were performed with non-labeled drug and high pressure liq. chromatog. anal. Following both oral and i.v. administration, the major route of elimination of drug-related compds. was via the feces for all species studied. Comparison of the oral and i.v. data shows that doxazosin is completely absorbed in man, rat, and mouse and is moderately well absorbed in dog. Doxazosin is extensively metabolized; only about 5% of the dose was excreted unchanged in man and main routes of metab. involve 6- and 7-O-demethylation and 6'- and 7'-hydroxylation; these and some minor products were common to the mouse, rat or dog and man. Oral bioavailability of doxazosin is high in man (63%) in comparison with dog (60%) and rat (50%). Mean plasma clearance values (mL/min/kg) range from 1.2 (man) through 13 (dog) to 30 (rat), while mean elimination half-life values (t1/2, hours) are 9 (man), 4.7 (dog) and 1.2 (rat). The long half-life in man provides the basis for once-daily dosing during doxazosin therapy. .
