DOCUMENT TYPE: Journal CA Section: 4 (Toxicology) Various factors affecting the indirect hemagglutination test for the titrn. of tetanus antitoxin were evaluated with a view to obtaining max. sensitivity in tests using unfixed sheep erythrocytes and sheep erythrocytes fixed with glutaraldehyde [111-30-8], formaldehyde [50-00-0], and pyruvic aldehyde [78-98-8]. The optimal concn. of tannic acid was 1/40,000 for tanning both fixed and unfixed sheep erythrocytes. Tanned sheep erythrocytes sensitized with 50 Lf/mL of tetanus toxoid at pH 7.2 for 1 h were the most sensitive. Although the optimal temp. of sensitization was 56°, unfixed cells tended to clump and lyse at this temp. Thus, a temp. of 37° was used to sensitize unfixed sheep erythrocytes. Sheep erythrocytes from different animals and the final concn. of sensitized sheep erythrocytes both had great effects on sensitivity. A final concn. of 0. 50-00-0 and 111-30-8 are just another two chemicals used in this study.5% of sensitized sheep erythrocytes was suitable as a compromise between sensitivity and readability. The loss of sensitivity of fixed and sensitized erythrocytes was investigated by storing these cells at 4-8° for 6-9 mo. .

By this method, mutagens such as glyoxal [107-22-2], methylglyoxal [78-98-8], 2-(2-furyl)-3-(5-nitrofuryl)acrylamide [3688-53-7], and 4-nitroquinoline N-oxide [56-57-5], used as model compds., were detected rapidly with high sensitivity. Reaction with isopropylideneguanosine (IPG)(II) [362-76-5], followed by isolation and characterization of the mutagen-IPG-adduct was a useful method for identifying unknown mutagens in crude samples. This method was successfully applied in identification of the mutagens in heated glucose (200°, 20 min); glyoxal-IPG [89780-74-5] and 8-hydroxy-IPG [13565-77-0] were identified in the reaction mixt. .
