DOCUMENT TYPE: Journal CA Section: 1 (Pharmacodynamics) Ten metabolites of Clofedanol (I) [791-35-5] were identified in human urine. The following 5 products were extd. with CHCl3 at pH 1-2: o-(chlorophenyl)phenylmethane [29921-41-3], benzophenone [119-61-9], o-chlorobenzophenone [5162-03-8], benzhydrol [91-01-0], and o-(chlorophenyl)-b-phenylacrolein [66171-83-3]. Two compds. were extd. with CHCl3 at pH 13-14: unchanged I and 1-(o-chlorophenyl)-1-phenyl-3-dimethylamino-1-propene [66171-84-4]. N,N-dimethylamino-acetic acid [1118-68-9] was also identified in urine. I and benzhydrol were present as free compds., and they were also found after hydrolysis of their resp. glucuronides with b-glucuronidase.

Tablets are dissolved in a H2O-MeCN () mixt. Samples are sepd. on an octadecylsilane column with a mobile phase of 70% MeCN and 30% aq. soln. of 0.005M 1-octanesulfonic acid, 1% HOAc, and 1% diethylamine. Effluent is monitored by a spectrophotometric detector with a 254-nm filter. I is sepd. from a complex mixt. of plant exts. and fillers in the tablets.
