DOCUMENT TYPE: Journal CA Section: 1 (Pharmacology) The development of 3 analogous radioimmunoassay procedures for dihydroergotoxine mesylate [8067-24-1] components is described. The antisera were produced by immunization of rabbits with immunogens obtained by coupling egg albumin to the indole group of each ergot alkaloid deriv. In each radioimmunoassay, antibodies do not cross-react more than 5% with the 2 other derivs. The tracers iodinated with 125I were prepd. by the chloramine-T method and purified by thin-layer chromatog. Both the high antibody affinity and high specific radioactivity of the tracers allow a sensitive assay (detection limit <20 pg/mL) in human plasma. After high-performance liq. chromatog. of the extd. plasma, no immunoreactive materials other than those corresponding to the elution of the 3 dihydroergotoxine components were detected. Two preliminary pharmacokinetic profiles obtained in dogs and humans for each deriv. are shown. .

This was true in rats with the addnl. risk factors of age and hypertension, as well as in young and normotensive rats.
