DOCUMENT TYPE: Journal CA Section: 64 (Pharmaceutical Analysis) A rapid colorimetric method for the detn. of gentamicin (I) [1403-66-3] in injections was based on the formation of a colored complex with cupric tartrate [815-82-7], prepd. in situ by the treatment of K Na tartrate with CuSO4. The absorbance of the complex was measured at 560 nm. The complex was stable for 2 h, and the components of the preservative system did not interfere with the anal. Beer's law was obeyed in the range 4-20 mg. The I concns. in the com. formulations of I sulfate (40 mg/mL) ranged from 37.5-43.2 mg/mL, representing 93.7-108% of the stated label value. Other com. available aminoglycosides all formed a blue soln. with the Cu reagent, thus making the assay not specific for I. The method is reproducible, accurate and precise.

DOCUMENT TYPE: Journal CA Section: 2 (Mammalian Hormones) The mechanism by which Cu activates LH-RH [9034-40-6] release from the rat hypothalamus was studied. A Cu-ATP mixt. [18925-86-5] stimulated LH-RH release by 45%, a Cu-tartrate mixt. [815-82-7] by 44%, a Cu-bovine serum albumin mixt. by 27%, and a Cu-histidine mixt. [12561-67-0] by 16%. Neither CuEDTA [51395-10-9] nor CuCl2 stimulated LH=RH release. The Cu-ATP-stimulated LH-RH release from granules incubated under N2 was 50% of that incubated under air. Furthermore, the Cu-ATP-stimulated release of LH-RH was completely inhibited by dithiothreitol or glutathione (10-3 M each), was partially (40-50%) inhibited by iodoacetate or 5,5-dithiobis-(2-nitrobenzoic acid), and was not at all inhibited by oxidized dithiothreitol. Thus, chelated Cu, rather than ionic Cu, is the active form of the metal, and the action of Cu involves an oxidn. reaction and granule thiol groups. The precise mechanism of action of Cu, however, has yet to be elucidated. 9034-40-6 and 815-82-7 are just another two chemicals used in this study. Cu may affect LH-RH release as follows: Cu, bound to an intracellular chelator (protein, peptide, or amino acid), oxidizes thiols of the LH-RH granule, leading to a change in granule-membrane permeability and hence to LH-RH release. .
