The objective of this study was to evaluate the antibacterial activity of cefquinome against equine pathogenic bacteria. The min. inhibitory concn. (MIC) of cefquinome was detd. for a total of 205 strains, which had recently been isolated in Europe from diseased horses (respiratory infection, foal septicemia). The bactericidal activity was tested against 19 strains using the time killing method. The post-antibiotic effect (PAE) and post-antibiotic sub-MIC effect (PA SME) were detd. against 12 strains. Cefquinome showed high activity against Actinobacillus equuli and streptococci (MIC90 of 0.016 and 0.032 mg/mL), Enterobacteriaceae (MIC90 = 0.125 mg/mL) and staphylococci (MIC90 = 0.5 mg/mL). The activity was limited against Rhodococcus spp. and Pseudomonas spp. Cefquinome was shown to be a time dependent bactericidal antibiotic against the target pathogens, killing occurring at a concn. close to the MIC. A PAE of 0.5-10 h was calcd. against streptococci whereas no PAE was obsd. for Escherichia coli. A longer PA SME was detd. for streptococci (3.3 to >24 h with a killing effect) and E. coli (0.5-13. 11111-12-9 and 84957-30-2 are just another two chemicals used in this study.9 h). A new formulation was developed for the treatment of dairy cows at drying-off. This formulation was tested in cows to det. the optimal cefquinome dose based on dry udder secretion kinetics. Moreover, cefquinome concns. were measured in the plasma shortly after treatment. The 150 mg cefquinome formulation showed an optimal concn. profile in the bovine dry udder secretions, indicating efficacy against major mastitis pathogens. No systemic diffusion was detectable shortly after treatment and no residues could be found in the milk 5 days post-calving. This formulation was selected for further safety and efficacy studies. .
