The anal. techniques uses plasma protein pptn. and direct injection on a C18 column, with an isocratic mobile phase and spectrofluorometric detection. 3R-Hydroxyquinidine was employed as the internal std. Linearity was verified up to 5 mg/L for the 2 drugs; concns. between 0.5 and 2.5 mg/L were measured with a coeff. of variation of 0.5-2.07% for a given day and a sensitivity limit of 50 mg/L. Plasma concn.-time profiles and pharmacokinetic parameters in 3 dogs were studied after i.v. or oral administration. A significant difference is obsd. in terminal half-life, terminal rate const., and total clearance of the 2 polar analog of quinidine. .
